NEUROFILAMENT LIGHT CHAIN LEVELS DURING RELAPSING PHASES OF MULTIPLE SCLEROSIS AND NEUROMIELITE OPTIC SPECTRUM DISORDER

Resumo

Introduction: Multiple Sclerosis (MS) and Neuromyelitis Optic Spectrum Disorders (NMOSD) are autoimmune diseases that present different morbidity. NMOSD patients reach higher levels of EDSS and lethality rate in a shorter interval of time when comparing with MS, due to more aggressive relapsing. Brazilian population is characterized by high miscegenation and ethnicity is also associated to disease severity. Serum Neurofilament light chain (NfL) level is a noninvasive toll to measure neurodegeneration.
Objective: Measuring Serum Neurofilament Light Chain level during relapsing phases of MS and NMOSD patients.
Methods: We include five of 60 NMOSD patients, nine of 250 MS patients during relapsing phase and 24 healthy control matched by age. Plasma samples were underwent by single molecule array (SIMOA) to investigate NfL level according to the manufacturer's instructions, with limit of detection 0.05 pg/mL.
Results: Among NMOSD patients, all were female with mean age of 44.4 years, and among MS patients six were female with mean age of 42.8 years. Healthy control mean age was 40.5 years. NfL level MS patients was 26.99 pg/mL, NfL level in NMOSD patients was 244.64 pg/mL and in healthy control was 6.77 pg/mL.
Conclusion: Significant difference between MS and NMOSD NfL levels (p < 0.01) seem to be in according to more severity pathophysiology of NMOSD and reinforce the necessity of aggressive treatment to prevent and treat autoimmune diseases associated to neurodegenerative process. Yet, our NfL results in Brazilian NMOSD patients can possible be associated to the population ethnicity and need to be repeated in a larger number of patients and population background.
