MICRORNA-205-5P E MICRORNA-130A-3P PROMOTED PROLIFERATION AND MIGRATION IN CERVICAL CANCER CELL LINES
Introdução
Genetics and epigenetics changes have been showing as crucial regulators for cervical cancer progression. In this context, emerging evidence indicates the microRNAs (miRNAs) play essential roles in tumorigenesis and development. Recent studies have shown that miR-205-5p and miR130a-3p could be used as good biomarkers for CIN 3. In addition, these miRNAs may be involved in the main stages of cervical cancer progression.
Objetivo
We aimed to evaluate the functional role of two miRNAs, miR-205-5p and miR-130a-3p, in a panel of cervical cancer cell lines.
Método
We used a cervical cancer cell lines panel (HeLa, SiHa, CasKi, C-4 I, C33-A, and HCB-514). The miRNA expression was evaluated by quantitative PCR (qPCR). We used a negative control A (scramble) and LNA miRNA inhibitor (miR-205-5p and miR-130a-3p inhibitor), and the capacity of proliferation, migration, and apoptosis in vitro for the CaSki strain was evaluated.
Resultado
The CaSki, C4-I, and HCB-514 cell lines showed overexpression of both miRNAs (miR-205-5p and miR-130a-3p), the C33-A cell lines showed overexpression only for miR-205-5p. The decreased expression levels of miRNAs, miR-205-5p and miR130a-3p, induced the migration of CaSki and C33-A cells, and also the proliferation in CaSki, but there was no significant difference in the suppression of apoptosis in these cells.
Conclusão
The miRNAs, miR-205-5, and miR-130a-3p, contribute to the proliferation and migration of CC cells and are biomarkers to identify CC progression and possible potential for the treatment of this neoplasia.
