Evaluation of NKCC1 gene expression in brain structures after administration of mesenchymal stem cells in a model of acute epilepsy

Introdução

Epilepsy is a condition characterized by the occurrence of spontaneous seizures and recurrent and synchronized bursts in the CNS. Temporal Lobe Epilepsy (TLE) is one of the most prevalent type of epilepsy and is characterized by rhythmic and synchronized firing of neuronal populations that lead to spontaneous and recurrent seizures. About 30% of individuals affected by this disturb do not respond to drug treatment. Therefore, it is necessary to search for new therapeutic for this disease. Mesenchymal stem cells (MSCs) have multipotent capacity and can differentiate into different tissues, in addition to the ability to modulate diseased niches.

Objetivo

Thus, the present work aimed demonstrate that MSCs can affect gene expression of cation-chloride co-transporter NKCC1 in the context the pilocarpine-induced TLE animal model.

Método

For this purpose, MSCs were extracted from the bone marrow of Wistar rats, cultured and transplanted into healthy Wistar rats and animals induced to status epilepticus by pilocarpine. The animals were separated into two groups: control and pilocarpine-induced status epilepticus, then they were euthanized 1 and 7-days post-transplant. MSCs were transplated via intranasal. Expression of NKCC1 were analyzed by RT-PCR.

Resultados

As a result, we were able to observe that the MSCs changed the amount of transcript in the brain of the animals. When gene expression was evaluated after intranasal administration, it was observed that amygdala, hippocampus and prefrontal cortex presented alteration. Hippocampus and prefrontal cortex altered gene expression both 1 and 7-days post-transplant. On the other hand, entorhinal cortex did not presented alteration on NKCC1 expression after MSCs instranasal administration, regardless of MSCs administration time.

Conclusão

These results show that MSCs can modulate gene expression of cation-chloride co-transporter NKCC1 during acute phase of epilepsy.
